- Veteran Moisés Duque will play for Os Cobras for the first time in 2025. Duque is one of seven changes to Os Cobras. Yacare have made just two alterations.
- Joaquin Lamas is serving suspension following his red card against Los Pampas.
- Os Cobras line-up contains 21 Brazilian eligible players. The two exceptions are Argentines Javier Coronel and Manuel Todaro.
- Yacare is majority Argentine; eight starting players are not eligible for Paraguay. They are Mariano Muntaner, Axel Zapata, Lucas Sommer, Felipe Puertas, Nicolás Heit, Juan Cruz Strada, Valentino DiCapua, and Julián Quetglas. Os Cobras are in search of their first win of Super Rugby Americas 2025. The team has been competitive at the set-piece but fallen short in other areas. They are up against a team who likes a hard battle in the forwards. Yacare ‘s two wins this season featured tries but it was the forwards who shone the brightest. The link has come from scrum-half Juan Cruz Strada who has been exceptional this season. Yacare are favorites to win away.
